ABSTRACT

OBJECTIVE To establish the fingerprints of Xanthoceras sorbifolia and determine the contents of flavonoids . METHODS HPLC was adopted . Using epigallocatechin as reference ,the fingerprints of 11 batches(No. S1-S11)of X. sorbifolia were drawn with Similarity Evaluation System of Chromatographic Fingerprints of TCM (2004A edition ). The similarity evaluation was conducted ,the common peaks were also confirmed . Cluster analysis (CA)and principal component analysis (PCA)were also performed. Epigallocatechin was selected as internal reference ,and quantitative analysis of multi -components by single marker (QAMS)was used to determine the contents of gallocatechin ,catechin,epicatechin,dihydromyricetin,taxifolin and myricetin in 16 batches(No. S1-S16)of X. sorbifolia. The results were compared with the results of one point external standard method and standard curve method . RESULTS There were 15 common peaks in 11 batches of X. sorbifolia,and the similarity of them were 0.910-1.000. A total of 7 common peaks were identified ,i.e. galliccatechin(peak 1),epigallocatechin(peak 2),catechin(peak 3),epicatechin(peak 5),dihydromyricetin(peak 6),taxifolin(peak 14)and myricetin (peak 15). The results of CA showed that S5-S7 and S 9 were clustered into one category ,S8 and S 11 were clustered into one category ,S10 were clustered into one category,S1-S4 were clustered into one category . The results of PCA showed that accumulative variance contribution rate of 3 principal components was 99.24%;S5-S7 were clustered into one category ,S8-S11 were clustered into one category ,S3 and S 4 were clustered into one category ,S1 and S 2 were clustered into one category . With the exception of myricetin and a partial batches (S12,S14-S16) of catechin ,the RSDs measured by the three methods for galliccatechin ,catechin (remaining batches ), epicatechin,dihydromyricetin and taxifolin in 16 batches of X. sorbifolia were less than 4% (n=3). CONCLUSIONS The established HPLC fingerprint and the method for content determination can be used for the quality control of X. sorbifolia. QAMS method can be used for the content determination of galliccatechin,epicatechin,dihydromyricetin and taxifolin .

ABSTRACT

OBJECTIVE@#To explore the molecular pathogenesis for a pedigree affected with hereditary coagulation factor XII (FXII) deficiency.@*METHODS@#Potential variant of the F12 gene was analyzed by PCR and Sanger sequencing. Expression plasmids were constructed by site-directed mutagenesis based on the wild-type and transiently transfected into 293T cells. FXII:C and FXII:Ag of the expression products were determined in the supernatant and cell lysate. Western blotting was used to verify the identify of the protein.@*RESULTS@#Gene sequencing revealed that the proband has carried 46TT genetype and heterozygous p.Glu502Lys variants in exon 13, and a heterozygous p.Gly542Ser variant in exon 14 of the F12 gene. Transfection experiment suggested that the FXII:C and FXII:Ag of p.Glu502Lys variant in the supernatant were 28% and 24%, compared with the wild-type (100%) and FXII:Ag of cell lysates was 39% compared to the wild-type (100%). The FXII:C and FXII:Ag of p. Gly542Ser variant in the supernatant were 32% and 17% and the FXII:Ag of cell lysates was 59%.@*CONCLUSION@#The 46TT genetype, p.Glu502Lys and p.Gly542Ser variants of the F12 gene probably underlie the low FXII level in the proband. As shown by in vitro experiment, the p.Glu502Lys and p.Gly542Ser variants can both inhibit the synthesis and secrection of the FXII protein.

ABSTRACT

Depression is a major mood disorder. Abnormal expression of glial glutamate transporter-1 (GLT-1) is associated with depression. Schisantherin B (STB) is one bioactive of lignans isolated from Schisandra chinensis (Turcz.) Baill which has been commonly used as a traditional herbal medicine for thousands of years. This paper was designed to investigate the effects of STB on depressive mice induced by forced swimming test (FST). Additionally, we also assessed the impairment of FST on cognitive function in mice with different ages. FST and open field test (OFT) were used for assessing depressive symptoms, and Y-maze was used for evaluating cognition processes. Our study showed that STB acting as an antidepressant, which increased GLT-1 levels by promoting PI3K/AKT/mTOR pathway. Although the damage is reversible, short-term learning and memory impairment caused by FST test is more serious in the aged mice, and STB also exerts cognition improvement ability in the meanwhile. Our findings suggested that STB might be a promising therapeutic agent of depression by regulating the GLT-1 restoration as well as activating PI3K/AKT/mTOR pathway.

ABSTRACT

Objective:To investigate the labial(buccal)bone thickness at alveolar crest zone and alveolar crest height of the maxil-lary anterior teeth and premolars of young adults with normal occlusion.Methods:The alveolar bone of the anterior teeth and premo-lars of 67 eligible Han national young volunteers was scanned by CBCT.Then the facial bone thickness and the distance between the facial alveolar crest and Cemento-enamel Junction(CEJ)of the anterior teeth and premolars were measured and analyzed after recon-struction.Results:The distance between labial (buccal)crest and labial (buccal)CEJ of the maxillary first premolars was the lar-gest(P <0.05);there was a negative correlation between the labial(buccal)crest height and the facial alveolar bone thickness at 2 mm from CEJ toward root derection[(P <0.05),0.6 <|r|<0.8].Conclusion:The labial (buccal)crest of the first premolars was higher than that of other teeth in maxillary aesthetic zone.At alveolar crest zone,when the labial (buccal)bone was thinner,the dis-tance between labial (buccal)crest and labial (buccal)CEJ was larger,and the implant aesthetic risk is higher.

ABSTRACT

Objective To optimize the extraction process of Radix Paeoniae Alba in Baijin Capsule by orthogonal experiment.Methods The study employed the extraction rate of paeoniflorin and total glucosides of paeony as evaluation indexes. The orthogonal design was used to investigate effects of solvent volume, extraction time and extraction frequency on extraction results.Results The optimal extracting condition was extracted 3 times, with 14 fold 70% alcohol, 1.5 h for each time. Conclusion The method is simple and steady, which will provide instruction and reference to the production of Baijin Capsule.
